Peptide Fun Fact: The first synthetic peptide (Gly-Gly) was produced by Emil Fischer in 1901 using unprotected amino acids [Ber. Dtsch. Chem. Ges. 34 (1901) 2868].  With the development of N-protected amino acids and methods to activate them for amide bond formation, the complexity of synthesized peptide targets increased. In 1953 Vincent du Vigneaud completed the total synthesis of the cyclic peptide oxytocin [J. Am. Chem. Soc. 76 (1954) 3115]. This effort was undertaken to characterize the structure of this hormone and led to his winning of the 1955 Nobel Prize in Chemistry. Another significant synthetic milestone in the history of biologically active peptides was the total synthesis of the enzyme ribonuclease A in 1969 by Gutte and Merrifield [J. Am. Chem. Soc. 91 (1969) 501]. This 124 residue enzyme was produced via solid phase synthesis using Boc protected amino acids and dicyclohexy!carbodiimide (DCC) in an overall yield of 17%. The work of Fischer, Vignead, and Merrifield helped lay the groundwork for our current ability to produce pharmaceutically active peptides on the kilogram scale.
